Separation conditions were studied on the analytical scale for the preparative separation of corosolic acid from plant extract. After separation conditions were established with 5 µm particle size, the particle size was changed to 10 µm which would be used for the actual preparative separation. Though resolution between the target compound and impurity decreased on changing to 10 µm packing material, sufficient resolution was obtained by reducing flow rate.

Loading was gradually increased, and purity and recovery rate of the preparative fractions were determined. The final loading was chosen based on target purity and recovery.
Based on the result of method development and loading study on the analytical scale, the large-scale preparative separation was performed.
